Abstract

<p>The most basic way to assess the phenotype of single-celled organisms is to measure their doubling time during the exponential phase of growth in liquid culture at the optimum growth temperature.This protocol is used to compare the doubling time (also known as generation time) of wild type yeast cells (Saccharomyces cerevisiae) with a gene deletion strain from the Yeast Deletion Collection to determine whether the cells carrying the mutation are impaired for growth at 30°C (the optimum growth temperature for yeast) and/or at 13°C (cold stress temperature).</p>
